If sounds like some kind of typo in grub.conf. If you can manually type
in the kernel line and the system boots, it has to be something in the
grub.conf file.
Maybe try deleteing the root and kernel lines, and retyping them (no cut
and paste). If that doesn't work, maybe post your grub.conf so we can
see it.
