I'm also quite new to this list (and gentoo programming). 
As far as I understood you, isn't this is what you're looking for:
DEPEND="tcltk? ( dev-lang/tcl dev-lang/tk)"

src_compile() {
    use tcltk \
        && myopts="${myopts} --enable-tcl" \
        || myopts="${myopts} --disable-tcl"
        #Or whatever options should enable or disable it.
